it's an action-rts. some call it a multiplayer online battle arena, but that term is extremely stupid in my opinion.
everyone starts the match level 1 with a certain amount of gold (i think it was 503? i'm a league player, i don't know). there are three "lanes" where AI-controlled "creeps" charge down. there are three towers in each lane, and behind the last tower are barracks, which affect the production of creeps (don't know what it does in dota). in between the lanes are jungles, narrow mazes with neutral creeps. inside every base is an ancient, and if the ancient dies then you lose the game. there are two more towers protecting the ancient. you just go to a lane (two top, one mid, two bot. or solo top, one mid, two bot, one jungle. or solo top, one mid, three bot), and focus on killing enemy creeps (getting the last hit) for gold and experience. you're not alone in the lane, there is also another enemy laning against you. when you die, you have to wait for a respawn, and you lose gold. you can buy items from the shop, ranging from a tree branch giving a little bit of everything for 53 gold, to an extremely valuable sword that really, really, really significantly increases your damage but is dropped on death (and can be picked up by enemies) for 5200 gold. that's the very basic of it, go look up "welcome to dota 2, you suck" by purge for a better explanation. |

and the difference between league and dota?
league:
-has cheap, low-cooldown, low-effectiveness abilities that generally need to be aimed (skillshots) that scale with stats (mainly ability power, some scale with attack damage though), allowing for mages to carry and snowball.
-is a bit more forgiving. you don't lose gold on death, you can't deny creeps (kill allied creeps to deny gold), and you can freely teleport back to base as long as you survive the 8 second channel time.
-is a bit more action-based. stuff like this: http://youtu.be/4PLgn_5mNxg or this: http://youtu.be/aiA5Tt1RHcg are impossible in dota. teamfights are decided on who can land and dodge abilities better. movement feels much smoother than dota, and abilities have, at most, a 0.25 second cast time. -balances around reworks. there are two champions being reworked next patch, for example.
-has four different maps: summoner's rift (default dota-style), dominion (5v5 control points), twisted treeline (3v3 dota-style), and howling abyss (5v5, one lane, random champions. it's fun.)
-relies on a f2p model based around you buying champions (they're locked unless you buy them or it's free week), and buying skins. tbh dota 2 has a better f2p model, but the skins are better than dota's equipment. compare one of the more basic skins: http://youtu.be/-WlRx6rNgp4 , and the ultimate skin: http://youtu.be/_RXjDym_cJE
dota:
-has high mana cost, high cooldown, high effectivness abilities that are generally targeted or aoe. they don't, however, scale with stats (they will always deal, say, 500 damage), which prevents mages from snowballing, and allowing for potential to shut down someone who is getting fed and carrying.
-is less forgiving, but is a much more complex game. you lose gold when you die, you need to pay 135 gold to teleport, you can deny creeps, and a lane can snowball out of control just from one tiny misstep. vision wards are also very limited.
-is a bit less action-based, and a bit more strategy-based. this is the most famous dota 2 play ever: http://youtu.be/La--egn0MrU?t=27m12s , and it wasn't extreme mechanics that made this happen, it was expecting this to happen, and coordinating with the rest of your team to counter it. na'vi knew about the naga siren pick, and picked a rubick + enigma + juggernaut combo to counter it, and built a black king bar on enigma. so, this teamfight was actually decided from the hero pick/ban phase.
-balances around buffs, buffs, buffs, buffs. every hero is overpowered, which forms a weird balance.
-has only one map, but the map has been fine-tuned to perfection and no two games are the same.
-relies on a f2p model where you have every single hero in the game unlocked (because dota is so counterpick-based), and following almost exactly like tf2's f2p style. |

good choices
twisted fate is difficult, but he's such a great champion in general. but he definitely is super difficult, to the point where i can't play him.
ezreal is fun, fun, fun, fun, and fun.
you either love or hate quinn. she used to be super weak, but now she's pretty strong right now, being a ridiculous lane bully with strong teamfight cleanup potential and splitpushing power. |

her ult is meant for only two things:
1. running away 2. chasing someone
the new abilities don't mean crap, the only thing that matters is the ridiculous movespeed. the "running away" part is why she's such a good splitpusher. |
